Structure of Cardiac T-Tubules

چکیده

action potential through the heart. During the cardiac action potential, Ca2+ enters each cardiac myocyte through L-type Ca2+ channels [as the Ca2+ current (ICa)] and activates adjacent Ca2+ release channels [ryanodine receptors (RyR)] in the membrane of the intracellular Ca2+ store, the sarcoplasmic reticulum (SR), causing Ca2+ release from the SR (5). Ventricular myocytes, which cause contraction of the ventricles of the heart, have invaginations of the surface membrane, called transverse (t-) tubules, which are important in ensuring rapid and synchronous contraction of the cell, and hence of the heart (13). This brief review will describe recent evidence that t-tubules are regions of the cell membrane that play a specialized role in transmembrane ion flux, and hence cell function, and that this role may change, and hence contribute to the altered function observed, during heart failure.

T-tubule function in mammalian cardiac myocytes.

The transverse tubules (t-tubules) of mammalian cardiac ventricular myocytes are invaginations of the surface membrane. Recent studies have suggested that the structure and function of the t-tubules are more complex than previously believed; in particular, many of the proteins involved in cellular Ca2+ cycling appear to be concentrated at the t-tubule. Thus, the t-tubules are an important deter...

The structure and function of cardiac t-tubules in health and disease.

The transverse tubules (t-tubules) are invaginations of the cell membrane rich in several ion channels and other proteins devoted to the critical task of excitation-contraction coupling in cardiac muscle cells (cardiomyocytes). They are thought to promote the synchronous activation of the whole depth of the cell despite the fact that the signal to contract is relayed across the external membran...

In situ single photon confocal imaging of cardiomyocyte T-tubule system from Langendorff-perfused hearts

Transverse tubules (T-tubules) are orderly invaginations of the sarcolemma in mammalian cardiomyocytes. The integrity of T-tubule architecture is critical for cardiac excitation-contraction coupling function. T-tubule remodeling is recognized as a key player in cardiac dysfunction. Early studies on T-tubule structure were based on electron microscopy, which uncovered important information about...

Localization of functional endothelin receptor signaling complexes in cardiac transverse tubules.

Endothelin-1 (ET-1) is an autocrine factor in the mammalian heart important in enhancing cardiac performance, protecting against myocardial ischemia, and initiating the development of cardiac hypertrophy. The ETA receptor is a seven-transmembrane G-protein-coupled receptor whose precise subcellular localization in cardiac muscle is unknown. Here we used fluorescein ET-1 and 125I-ET-1 to provide...

Ultra Structure of Excretory Organ (Kidney) in Juvenile Grouper, Epinephelus coioides

In order to study of kidney tubules, twenty specimen of 45 days old juvenile grouper (Epinephelus coioides) were prepared after proper biometry. Five mm thick sections were removed from the middle and posterior end of the kidney and fixed in Bouin and glutaraldehyde. The routine procedures of preparation of tissues were followed for light and transmission electron microscopic study. In macrosco...

Dystrophin-glycoprotein complex and laminin colocalize to the sarcolemma and transverse tubules of cardiac muscle.

The expression and subcellular distribution of the dystrophin-glycoprotein complex and laminin were examined in cardiac muscle by immunoblot and immunofluorescence analysis of rabbit and sheep papillary muscle. The five dystrophin-associated proteins (DAPs), 156-DAG, 59-DAP, 50-DAG, 43-DAG, and 35-DAG, were identified in rabbit ventricular muscle and found to codistribute with dystrophin in bot...
